What is Daridorexant?
Daridorexant is an active substance belonging to a class of medicines known as orexin receptor antagonists.
Daridorexant treats the sleep disorder insomnia that is defined as difficulty falling asleep and/or staying asleep, despite enough time and ideal conditions. Insomnia causes an inability to function normally during the day, sleepiness and problems concentrating, and can have long-term health effects.
Orexin is a natural chemical produced by nerve cells in the brain that keeps people awake. Daridorexant works by preventing orexin binding to orexin receptors, which blocks orexin from working and thus suppresses wakefulness. The medicine enables a person to fall asleep faster and remain asleep for longer, as well as improve their daytime functioning. Daridorexant was designed to provide most action during the night and have low activity the next morning.
How to use Daridorexant
Daridorexant is a tablet that comes in two strengths 25 mg and 50 mg. The usual dose is one 50 mg tablet per night. However, you will have to take the lower 25 mg tablet if you have a liver disorder or have to take other specific medicines. Daridorexant is taken at night, with water, 30 minutes before going to bed. Tablets should be taken for only as long as they are needed, otherwise your doctor will assess you after three months and regularly thereafter. The effects of the medicine may be closely monitored in people with certain conditions.
Taking the tablet with food or immediately after a meal does not stop the tablet from working, although it may take longer to take effect. Grapefruit or grapefruit juice may increase the blood daridorexant level if taken the evening, and alcohol can increase the possibility of poor balance and coordination.
Seek immediate medical attention if you have taken more daridorexant than recommended - this may increase sleepiness, weakness in muscle, and morning drowsiness. If you forget to take the tablet when going to bed, do not take it later into the night as you may experience morning drowsiness.
Who can use Daridorexant?
Daridorexant is for adults aged 18 years and older who have experienced insomnia symptoms for three months or longer and their ability to function during the day is considerably affected.
Daridorexant should not be taken by children and adolescents, and adults who are allergic to daridorexant or any of the tablet's other ingredients, or adults who have narcolepsy (i.e., they suddenly fall asleep at any time).
Discuss taking daridorexant with a doctor before starting treatment if you have/ever had suicidal thoughts; have depression, a psychiatric disorder, liver problems, breathing difficulties, a history of falls, and/or are over 65 years of age; take medicines affecting the brain (e.g., for depression or anxiety); and/or take/have taken recreational drugs or been addicted to drugs and/or alcohol.
All medicines, including those currently taken, recently taken, or might be taken, must be discussed with a doctor before daridorexant is used. Daridorexant may interact with some medicines that work in the brain, or treat blood coagulation disorders and cardiac impairment so take advice from a doctor. Additionally, people taking certain antifungal agents, antibiotics, immunosuppressants, HIV treatments, and cancer treatments should contact their doctor before starting daridorexant, as some of these medicines can alter the blood daridorexant level.
Daridorexant side effects
Immediately contact a doctor if you have depression and it worsens, or you think of harming yourself. A doctor should be informed if, on waking or on falling asleep, you are unable to move or talk (sleep paralysis) for several minutes or you hear or see disturbing images that are unreal (hallucinate). One in a hundred people may experience these effects. One in ten people experience excessive sleepiness, headache, tiredness, dizziness, and/or feeling sick.
